The 22/250 makes no sense at all to me, given you have a .243 - which will do everything and better. But prairie dog shooting can be hot and you want a gun that you can keep shooting. A 6BR is a great choice, though buying 1,000 pieces of brass for an extended shoot costs a bit to a lot more than a .20 Practical or a .22 Nosler - plus barrel heating and recoil is more pronounced. In my opinion - the .20 Practical (throated for 32-40 grain bullets) doesn't give you an honest 150 yard dramatic improvement over the .223 in real field conditions, though it is an improvement, especially if you are using light varmint bullets in the .223. The .22 Nosler, using high-BC bullets does great, while still keeping moderate barrel heat and recoil about the same as a .223. I think it is a better option than the BR for these reasons, as well. In my case, I shoot a .20 Practical and a .223 the most - alternating for shots under around 400 yards. Beyond that, I use the .22 Nosler until the shots are so far out that the .243 or 6 BR are called into play.
