I tried this and had about .0015 runout. When pre-drilling and then boring, runout has never been that high....zero to .0002.Wouldn’t using the finishing reamer from start to finish be the most accurate way?

Don't drill the whole thing at once, measure from the neck shoulder junction to the middle of the bushing and drill that amount. That way you have your pilot engaged. If your tailstock is centered you don't really need a pilot, but it helps with the confidence thing to start with one.Thanks for all the advice guys. Not doing enough to ruin my reamers since I’m only building for myself. I’m still no understanding how a reamer pilot can follow the rifling if it is pre bored. I’ll read more
Have always used finish reamers start to finish with an oil flush system, NO runout!

I use a test indicator just inside the chamber mouth, I agree with you that the OD at the base is probably larger than the reamer but believe it is so minimal that it isn't a problem.
