New Barrel, when to ladder test?

I've got a brand new 6.5x47L barrel. This will be my first attempt at a ladder test, at which I will conduct at 300yds. First I plan on breaking in the barrel with the following approach: 1 shot, clean - 2 shots, clean - 3 shots, clean - that's it. I'm just wondering if I can do the ladder test at this time to search for node's? Say I find a couple of nodes to work with, I'm just wondering if these nodes will change as I get a few more rounds through it? I just want to try to find a good node while shooting up the least amount of ammo. I ended up shooting up 250 rounds on my last build searching for nodes by shooting groups, and I was hoping the ladder test would eliminate some of that.
 
First, barrel break in. The barrel is broken in when I quits copper fouling. My last Krieger I did shoot and clean for 2 shots only, and started load development, it shoots like a house on fire. Some have taken up to 10 shots.

About ladder test, you can do one right after barrel break in, that seems to work for me.
