I've got a 22 bra that I'm getting a sample of Alpha 6 bra brass for. The reamer specs says it has a .254 neck but it has been reground once, so I'm not 100% sure of neck diameter. Anyway, I'll be necking down and turning. Is there a difference in necking down 1st then turning or turning as a 6mm then neck down?

Turn the necks after you neck it down to 22.
I'd neck down, then turn. If you turn first, you don't really know how much to turn. Also, if you turn first, then the lower part of the 6mm neck becomes thinner and it becomes part of your shoulder! Turn second and you know how much is needed and you know where to stop turning.
